⚙️ Principle

Simmons Citrate Agar tests the ability of microorganisms to use citrate as the sole carbon source and ammonium salts as the nitrogen source. Organisms that can utilize citrate produce alkaline byproducts, increasing the pH of the medium.

The pH indicator bromothymol blue changes color from green to blue, indicating a positive result. If citrate is not utilized, the medium remains green with no growth.

Product Description

SIMMONS CITRATE AGAR – HX-MEDIA

Product Overview

Simmons Citrate Agar is a highly specialized selective and differential culture medium used for the biochemical characterization of microorganisms based on their ability to utilize citrate as the sole carbon source and ammonium salts as the sole nitrogen source. It is one of the most widely employed media in microbiology laboratories for the differentiation of Gram-negative enteric bacilli, especially within the Enterobacteriaceae family, and plays a crucial role in routine diagnostic as well as research applications.

The medium is formulated according to Simmons’ modification of Koser’s citrate medium, improving its stability, clarity, and reliability for laboratory use. The selectivity of the medium is achieved through its minimal nutrient composition. Sodium citrate acts as the exclusive carbon source, while ammonium dihydrogen phosphate serves as the nitrogen source. This restrictive formulation ensures that only microorganisms possessing the necessary transport mechanisms and enzymatic systems—particularly citrate permease—are capable of metabolizing citrate and sustaining growth.

Simmons a key component of the medium is the pH indicator bromothymol blue, which provides a clear and distinct visual indication of metabolic activity. Initially, the medium has a neutral to slightly alkaline pH (approximately 6.8–7.0) and appears green in color. When citrate-positive organisms metabolize citrate, they convert ammonium salts into ammonia and ammonium hydroxide, resulting in an increase in pH (alkaline shift). This change causes the bromothymol blue indicator to shift from green to an intense Prussian blue, signifying a positive reaction. In contrast, organisms that cannot utilize citrate fail to grow, and the medium remains green, indicating a negative result.

Simmons Citrate Agar is typically dispensed as slants in test tubes, which provide an increased surface area and adequate aeration required for optimal growth of aerobic citrate-utilizing bacteria. The slanted surface also allows for better observation of both growth and color change. Care must be taken to perform light inoculation using a straight wire rather than a loop, as heavy inoculation or carryover of nutrients from enriched media may lead to false-positive results.

This medium is an integral component of the IMViC test series (Indole, Methyl Red, Voges-Proskauer, and Citrate), which is extensively used for the identification and differentiation of coliform bacteria. For example, Escherichia coli typically gives a citrate-negative result (green), whereas organisms like Klebsiella pneumoniae and Enterobacter aerogenes are citrate-positive (blue).

Simmons Citrate Agar is widely applied in:

  • Clinical microbiology laboratories for pathogen identification
  • Food and dairy testing to detect contamination by enteric bacteria
  • Water quality analysis for coliform differentiation
  • Pharmaceutical and environmental microbiology
  • Academic and research laboratories for teaching biochemical testing methods

The medium is known for its high reproducibility, stability, and ease of interpretation, making it a dependable choice for microbiologists. For optimal performance, it should be stored in a cool, dry place (10–30°C), protected from moisture and direct sunlight, and used before the expiry date to ensure accurate results.

Overall, Simmons Citrate Agar remains an essential diagnostic medium that provides clear biochemical differentiation, supporting accurate microbial identification and contributing significantly to microbiological quality control and research.
