Principle

Mueller Kauffmann Tetrathionate Broth Base works on the principle of selective enrichment of Salmonella species from mixed microbial populations. After the addition of iodine-iodide solution, sodium thiosulfate in the medium reacts to form tetrathionate, which inhibits many coliforms and competing enteric bacteria.

Salmonella species possess the enzyme tetrathionate reductase, enabling them to survive and multiply in the selective environment. Bile salts and brilliant green further suppress unwanted Gram-positive and Gram-negative organisms, increasing the selectivity of the medium.

Peptones provide essential nutrients, amino acids, vitamins, and nitrogen sources required for bacterial growth, while calcium carbonate neutralizes toxic metabolites formed during incubation and helps maintain favorable growth conditions.

Following incubation, enriched cultures are subcultured onto selective agar media for isolation and identification of Salmonella species.

🛠️ How to Use

  1. Suspend the required quantity of medium in distilled water.
  2. Heat gently with mixing to dissolve completely.
  3. Sterilize as recommended.
  4. Cool the medium and add sterile iodine-iodide solution before use.

  6. Incubate at recommended temperature for 18–24 hours.
  7. Subculture onto selective agar media for isolation and identification of Salmonella.
