The principle of Rappaport Vassiliadis (RV) Broth is based on selective enrichment under conditions that favor the growth of Salmonella while inhibiting competing microorganisms.
The medium incorporates:
High magnesium chloride concentration, creating elevated osmotic pressure that inhibits many non-target bacteria
Malachite green, which suppresses Gram-positive and many Gram-negative organisms
Low pH (~5.2), further restricting the growth of competing flora
Salmonella species are relatively resistant to these harsh conditions and can survive and multiply, whereas most other bacteria are inhibited.
When incubated (commonly at 41–43°C), the selective conditions further enhance the growth advantage of Salmonella, leading to its preferential enrichment, even when present in low numbers or in stressed conditions.
This principle makes RV Broth highly effective for selective recovery and detection of Salmonella from complex samples.

The medium is designed with selective components such as malachite green and high magnesium chloride concentration, along with a low pH (~5.2), which collectively inhibit the growth of competing microorganisms while favoring the growth of Salmonella. These conditions exploit the ability of Salmonella to survive under osmotic stress and selective environments.
RV Broth is typically used as a secondary enrichment medium following pre-enrichment (e.g., in buffered peptone water), significantly improving the recovery of low numbers of stressed or injured Salmonella cells.
It is widely used in food safety testing, poultry and dairy industries, environmental monitoring, water testing, and clinical microbiology laboratories. The medium enhances sensitivity, selectivity, and detection efficiency, making it an essential tool in Salmonella surveillance and quality control programs.

Prepare the broth and inoculate the sample.
Incubate at 41.5–43°C.
