Azospirillum Medium (without agar) is a nitrogen-free liquid enrichment medium formulated for the selective isolation and cultivation of microaerophilic, nitrogen-fixing bacteria, particularly Azospirillum species from soil, rhizosphere, and plant root samples.
Azospirillum Medium is intentionally devoid of combined nitrogen, ensuring that only microorganisms capable of fixing atmospheric nitrogen can grow. It is enriched with a carbon source such as malate, along with essential mineral salts and trace elements that support metabolic activity and energy generation required for nitrogen fixation.
Under suitable incubation conditions, Azospirillum Medium exhibits a distinctive subsurface pellicle formation due to its microaerophilic nature, which serves as a key indicator for its presence and growth. The medium supports efficient enrichment even when the organism is present in low numbers.
