Buffered Peptone Water A1 Quality – HX-Media

Buffered Peptone Water A1 Quality – HX-Media

Price range: ₹570.00 through ₹49,760.00

⚙️ Principle (Detailed)

Buffered Peptone Water functions based on resuscitation and stabilization of injured microorganisms:

  • Peptone
Provides readily available nutrients (amino acids, peptides, nitrogen) that support cellular repair and metabolic recovery
  • Phosphate Buffer System (Disodium & Monopotassium Phosphate)
Maintains a stable pH environment, preventing acidification during microbial metabolism
  • Sodium Chloride (if present)
Maintains osmotic balance, supporting cell integrity

Mechanism:

Sub-lethally injured cells recover in a non-stressful environment, regain metabolic activity, and multiply to detectable levels before being exposed to selective media.

📦 Available Packaging Options

    • 100 g Bottle (makes ~1.5–2 liters of medium)

    • 500 g Lab Pack

    • 1 kg Bulk Pack
(Custom sizes available depending on supplier.)

Product Description

Buffered Peptone Water – HX-Media

Product Overview

Buffered Peptone Water (BPW) is a non-selective pre-enrichment medium specifically formulated for the resuscitation and recovery of sub-lethally injured microorganisms, particularly enteric pathogens such as Salmonella spp., from food, water, environmental, and pharmaceutical samples.

Buffered Peptone Water plays a critical role in microbiological testing workflows, especially in food safety and regulatory testing, where microorganisms may be stressed or damaged due to processing conditions such as heat treatment, freezing, dehydration, preservatives, or pH changes. In such cases, direct inoculation onto selective media may fail to recover these injured cells. Buffered Peptone Water provides a gentle, nutrient-rich, and non-inhibitory environment, allowing microorganisms to repair cellular damage and regain viability before exposure to selective conditions.

The medium consists of peptone as a nutrient source and a phosphate buffer system, which maintains a stable pH (~7.2) during incubation. This buffering capacity is essential, as metabolic activity during recovery may produce acidic by-products that could otherwise inhibit bacterial repair and growth.

Buffered Peptone Water is widely recommended in international standard methods (ISO, FDA, BAM, APHA) as the primary pre-enrichment step in the detection of Salmonella and other enteric pathogens. After incubation in Buffered Peptone Water, samples are typically transferred to selective enrichment broths (e.g., Rappaport-Vassiliadis, Selenite Cystine) and subsequently plated on selective agars for isolation.

Due to its non-selective and protective nature, Buffered Peptone Water ensures maximum recovery of viable microorganisms, improving the sensitivity and reliability of microbiological testing procedures.

🔬 Key Features

  • Non-selective pre-enrichment medium
  • Promotes recovery of injured and stressed microorganisms
  • Essential for Salmonella detection protocols
  • Maintains stable pH via buffering system
  • Improves accuracy and sensitivity of testing
  • Compatible with ISO, FDA, BAM methods
  • Easy to prepare and use

🎯 Applications

  • Pre-enrichment for Salmonella detection
  • Food microbiology (meat, poultry, dairy, processed foods)
  • Water and environmental testing
  • Pharmaceutical microbiological quality control
  • Recovery of heat-injured or stressed bacteria
  • Sample preparation before selective enrichment

🧪 Composition (Typical)

  • Peptone : 10.0g
  • Sodium Chloride : 5.0g
  • Disodium Phosphate : 3.5g
  • Monopotassium Phosphate : 1.5g
  • Final pH: 7.2 ± 0.2 at 25°C

🛠️ How to Use

  1. Suspend the required quantity of BPW in distilled or purified water.
  2. Mix thoroughly to dissolve completely.
  3. Sterilize by autoclaving at 121°C for 15 minutes.
  4. Cool to room temperature.
  5. Add sample (typically 1:10 dilution, e.g., 25 g sample in 225 mL BPW).
  6. Incubate at 35–37°C for 18–24 hours.
  7. After incubation, transfer aliquots to selective enrichment media for further analysis.

Storage and Shelf Life

  • Store dehydrated medium at 10–30°C, dry conditions
  • Keep container tightly closed
  • Prepared medium: store at 2–8°C for short-term use
  • Shelf life: 2–3 years (dehydrated form)
