🧪 Composition (Typical)

  • Peptone – 10.0 g

🛠️ How to Use

  1. Suspend ~40 g of Sheep Blood Agar Base in 1 liter distilled water
  2. Heat with agitation to completely dissolve the medium
  3. Sterilize by autoclaving at 121°C for 15 minutes
  4. Cool the medium to 45–50°C (critical step)
  5. Add 5–10% sterile defibrinated sheep blood aseptically
  6. Mix gently to avoid air bubbles or hemolysis of RBCs
  7. Pour into sterile Petri plates and allow to solidify
  8. Store plates at 2–8°C until use

Storage and Shelf Life

  • Store at 15–25°C, dry and tightly sealed.
  • Protect from moisture, heat, and light.
