⚙️ Principle

The principle of EMB Agar is based on lactose fermentation and dye interaction:

  • Strong lactose fermenters such as Escherichia coli produce large amounts of acid, leading to the formation of dark colonies with a characteristic metallic green sheen
  • Moderate lactose fermenters produce pink to purple colonies
  • Non-lactose fermenters produce colorless or translucent colonies

The dyes eosin Y and methylene blue react with acid produced during lactose fermentation, forming colored complexes. At the same time, these dyes inhibit Gram-positive organisms, enhancing the selectivity of the medium.

  • Dipotassium phosphate : 2.0
  • Eosin Y : 0.4
  • Methylene blue : 0.065
  • Agar : 15.0
  • Final pH : 7.1 ± 0.2 at 25°C

🛠️ How to Use

  1. Suspend the required amount of medium in distilled water.
  2. Heat with constant stirring to dissolve completely.
  3. Sterilize by autoclaving at 121°C for 15 minutes.
  4. Cool to 45–50°C and pour into sterile Petri plates.
  5. Allow to solidify and inoculate aseptically.
  6. Incubate at 35–37°C for 18–24 hours.
  7. Observe colony morphology and color for identification.

Storage and Shelf Life

  • Store dehydrated medium at 10–30°C in a dry place
  • Keep container tightly closed and protected from light
  • Prepared plates should be stored at 2–8°C
  • Shelf life: 2–3 years under proper storage conditions
