The principle of Plate Count Agar is based on the growth and enumeration of viable microorganisms present in a sample under suitable nutrient and incubation conditions.
Microorganisms in the sample utilize the nutrients (tryptone, glucose, and yeast extract) and form visible colonies upon incubation. Each colony is assumed to arise from a single viable cell or group of cells (colony-forming unit, CFU).
By counting the number of colonies and considering the dilution factor, the total viable microbial count in the sample can be calculated, providing an estimate of microbial contamination or quality.

Plate Count Agar (PCA), also known as Tryptone Glucose Yeast Extract Agar, is a general-purpose, non-selective culture medium used for the enumeration of viable microorganisms in food, water, dairy, and environmental samples. It supports the growth of a wide range of heterotrophic bacteria, making it ideal for determining the total microbial load.
The medium contains tryptone as a source of amino acids, glucose as an ऊर्जा source, and yeast extract providing vitamins and growth factors. Its balanced formulation promotes consistent colony development, allowing accurate counting of microbial populations.
Plate Count Agar is widely used in food safety testing, dairy industries, water analysis, pharmaceutical quality control, and research laboratories, especially for standard plate count (SPC) or aerobic plate count (APC) methods.

Yeast Extract: 2.5 g
Glucose: 1.0 g
Agar: 15.0 g
Final pH: ~7.0 ± 0.2 at 25°C
Dissolve medium, sterilize at 121°C for 15 min.
Cool, pour plates, inoculate sample.
Incubate 30–37°C for 24–48 hrs and count colonies.
Store powder at 2–8°C, dry and protected from light.
Use freshly prepared plates; store at 2–8°C and use within a few days.
