Product Overview

Sulphate Reducing Medium (Twin Pack) is a specialized, high-performance culture medium system designed for the selective detection, enrichment, and cultivation of sulphate-reducing bacteria (SRB), a group of strict anaerobic microorganisms responsible for reducing sulphates to sulfides. These bacteria, including genera such as Desulfovibrio, Desulfotomaculum, and Desulfobacter, are widely encountered in industrial water systems, oil reservoirs, marine environments, and soil ecosystems.

The twin pack configuration consists of two separate components—typically a basal nutrient medium and a reducing/indicator supplement—which are combined just prior to use. This format ensures improved stability of sensitive ingredients, particularly reducing agents and iron salts, while also maintaining optimal redox conditions required for SRB growth. It also minimizes degradation during storage and enhances the overall reliability and reproducibility of results.

This medium is specifically formulated to support anaerobic metabolism, providing suitable carbon sources (such as lactate), essential nutrients, sulphate ions, and reducing agents that lower the oxidation-reduction potential of the medium. 🛠️ How to Use

  1. Prepare the basal medium by dissolving in distilled water
  2. Sterilize by autoclaving at 121°C for 15 minutes
  3. Cool to at 45–50°C
  4. Aseptically add the second component (reducing/indicator supplement)
  5. Mix gently to avoid oxygen incorporation
  6. Dispense into sterile tubes or bottles (preferably filled to minimize air space)
  7. Inoculate sample under strict aseptic and anaerobic conditions
  8. Incubate at 30–37°C for 3–7 days (or longer if required)
  9. Observe for black precipitate formation, indicating SRB growth

Storage and Shelf Life

  • Store both components at 10–30°C in a dry, well-ventilated area
  • Protect from moisture, heat, and direct sunlight
  • Keep containers tightly sealed
  • Reconstituted medium should be used immediately or within a short time
  • Shelf life: Typically 2–3 years under recommended conditions
