Pikovskaya’s Broth is a specialized microbiological medium formulated for the isolation, cultivation, and enumeration of phosphate-solubilizing microorganisms (PSM), particularly bacteria and fungi present in soil.

This medium is widely used in agricultural microbiology and environmental studies to identify microorganisms capable of converting insoluble phosphates into soluble forms, thereby enhancing soil fertility and plant growth. It is especially useful for screening biofertilizer strains such as Bacillus, Pseudomonas, and certain fungal species.

Pikovskaya’s Broth contains insoluble phosphate sources and essential nutrients that promote microbial growth while enabling the detection of phosphate solubilization activity through changes in the medium, such as increased turbidity or chemical analysis of soluble phosphate.

🧪 Composition (Typical)

• Glucose: ~10.0 g

• Tricalcium phosphate: ~5.0 g

• Ammonium sulfate: ~0.5 g

• Sodium chloride: ~0.2 g

• Potassium chloride: ~0.2 g

• Magnesium sulfate: ~0.1 g

• Yeast extract: ~0.5 g

• Manganese sulfate: trace

• Ferrous sulfate: trace

Final pH: ~7.0 ± 0.2

🛠️ How to Use

1. Suspend the required amount of Pikovskaya’s Broth powder in distilled water (as per label instructions).

2. Heat with gentle stirring to dissolve completely.

3. Dispense into flasks or tubes.

4. Sterilize by autoclaving at 121°C for 15 minutes.

5. Allow the medium to cool.

6. Inoculate with the test sample or culture.

7. Incubate at 28–30°C for 3–7 days or as required.

8. Analyze for phosphate solubilization through turbidity or chemical estimation of soluble phosphate.
