This medium works on the principle of selective fungal growth and bacterial inhibition:
Dextrose provides an abundant carbon and energy source for fungal growth.
Peptones supply nitrogen, amino acids, and growth factors.
Low pH (~5.6) inhibits many bacterial species while supporting fungi.
Chloramphenicol suppresses bacterial protein synthesis, preventing contamination.
This combination allows fungi to grow selectively in the broth, often indicated by turbidity or pellicle formation.

Sabouraud Dextrose Broth with Chloramphenicol is a selective liquid culture medium formulated for the cultivation and enrichment of yeasts and molds while inhibiting bacterial contamination.
This medium is based on Sabouraud Dextrose formulation, which contains a high concentration of dextrose and an acidic pH that favors fungal growth. The addition of chloramphenicol, a broad-spectrum antibiotic, enhances selectivity by suppressing the growth of most Gram-positive and Gram-negative bacteria.
It is widely used in clinical, pharmaceutical, food, and environmental microbiology laboratories for the isolation, cultivation, and maintenance of fungal organisms, including dermatophytes, Candida species, and other pathogenic fungi.

Dextrose: ~40.0 g
Peptone: ~10.0 g
Chloramphenicol: ~0.05 g (50 mg)
Final pH: ~5.6 ± 0.2 at 25°C
Suspend the required amount of medium in distilled water (as per label instructions).
Heat gently with stirring to dissolve completely.
Dispense into suitable containers.
Sterilize by autoclaving at 121°C for 15 minutes.
Cool to about 45–50°C (if antibiotic is added separately, add aseptically at this stage).
Inoculate the sample under aseptic conditions.
Incubate at 25–30°C for 2–5 days or longer depending on organism growth.
Store dehydrated medium at 10–30°C in a dry place
Protect from light and moisture (antibiotic is light-sensitive)
Keep container tightly closed
Prepared medium should be used fresh or stored at 2–8°C for short-term use
Shelf life: 2–3 years under recommended storage conditions
